To make sure your domain name remains registered to you and usable for your site make sure the renewal option is set for automatic and that your payment option is always up to date - such as keeping your credit card's expiration date current. Make certain your contact information at the registrar is current and especially that your email address on file with them is current. Also make sure to configure your email client to accept email messages from your registrar. Keep all passwords confidential.
Once you choose hosting for your new site you will need to log into your domain name registrar account and update the nameservers to point to your site. For instance, if you wish to have me build and host your site with my hosting service you would need to change your domain name's dns from free parking to ns1.sitewryte.com and ns2.sitewryte.com. But, if you will be hosting your site somewhere else you would get the nameserver information from the hosting service that you sign up with.
